RESUMO

The purpose of this study was to investigate the relationship between cyclin D1 expression and clinicopathological parameters in patients with prostate carcinoma. We assessed cyclin D1 expression by conventional immunohistochemistry in 85 patients who underwent radical prostatectomy for prostate carcinoma and 10 normal prostate tissue samples retrieved from autopsies. We measured nuclear immunostaining in the entire tumor area and based the results on the percentage of positive tumor cells. The preoperative prostate-specific antigen (PSA) level was 8.68±5.16 ng/mL (mean±SD). Cyclin D1 staining was positive (cyclin D1 expression in REPLACE_GT5% of tumor cells) in 64 cases (75.4%) and negative (cyclin D1 expression in ≤5% of tumor cells) in 21 cases (including 15 cases with no immunostaining). Normal prostate tissues were negative for cyclin D1. Among patients with a high-grade Gleason score (≧7), 86% of patients demonstrated cyclin D1 immunostaining of REPLACE_GT5% (PREPLACE_LT0.05). In the crude analysis of cyclin D1 expression, the high-grade Gleason score group showed a mean expression of 39.6%, compared to 26.9% in the low-grade Gleason score group (PREPLACE_LT0.05). Perineural invasion tended to be associated with cyclin D1 expression (P=0.07), whereas cyclin D1 expression was not associated with PSA levels or other parameters. Our results suggest that high cyclin D1 expression could be a potential marker for tumor aggressiveness.

A patologia descritiva das lesões em animais domésticos de açougue permite enriquecer a literatura médica veterinária especializada em inspeção sanitária de carnes, assim como subsidiar tecnicamente os profissionais inspetores. A oncocercose é uma parasitose causada pelo nematódeo Onchocerca sp,, sendo que a espécie O. gutturosa é a que mais acomete bovinos na América do Sul. Foram avaliados, pelo exame anatomopatológico, dois casos de lesões localizadas no ligamento nucal de bovinos abatidos para o consumo. A macroscopia de um caso revelou bursite crônica, representada por grande quantidade de líquido citrino e viscoso e, ainda, muitos corpúsculos livres, elípticos e achatados desprovidos de raiz (arrizo). A microscopia dessas estruturas evidenciou moldes de fibrinas amorfos e eosinofílicos. O segundo caso foi caracterizado por bursite aguda, e a microscopia das lesões ligamentar e periligamentar revelou granulomas epitelioides com reação gigantocitária, centralizados por formações semelhantes ao Onchocerca sp...


The descriptive pathology of lesions in butcher's domestic animals contribute to enrich literature specialized in veterinary sanitary inspection of meat as well as to guide professional meat inspectors. Oncorcecose is a parasitic disease caused by the nematode Onchocerca sp, and the O. gutturosa species affects cattle in South America. Two cases of lesions located in the nuchal ligament of cattle slaughtered for consumption were evaluated by pathological examination. Macroscopic analysis revealed a case of chronic bursitis represented by a large amount of clear and viscous liquid, and also many free, elliptical and flat corpuscles. Microscopy showed arrizoides molds fibrin, amorphous, and eosinophilic structures. The second case was characterized by acute bursitis and microscopic lesions and ligament periligamentous revealed multiple epithelioid granulomas and giant cells centered on formations similar to Onchocerca sp...

For several decades, the bivalve mollusk Anomalocardia brasiliana has been extracted in estuarine areas in the semi-arid region of northeastern Brazil. However, information on both dynamics and abundance of their populations are missing. In this context, the present study, therefore, investigated several aspects of the population structure of A. brasiliana on beaches near the estuary of the Apodi River in Northeast Brazil. The aims were to determine the population density of A. brasiliana during different times of the year, to estimate population parameters, and to relate species density and distribution to the abiotic characteristics of the region. Sampling was performed from March/2007 and May/2008, we collected samples of sediments and mollusks along 180 m transects in the intertidal area on the beaches Barra and Pernambuquinho, Grossos - RN. At each sampling site, samples of sediments and mollusks were collected in the intertidal area along 180 m transects. The highest and lowest average densities on Pernambuquinho beach were 1148 (April/2007), and 100 individuals.m2 (May/ 2008), respectively. On Barra beach, the highest and lowest densities were 1813 (April/2007) and 951 individuals.m2, (November/2007), respectively. The densities of A. brasiliana on both beaches were significantly different only in the months of January, April and May/ 2008. Length of the individuals ranged from 1 to 28 mm, with most individuals measuring between 22 and 24 mm. Length growth rate parameters for A. brasiliana were L = 28.68 mm and k = 0.61 year1 on Barra beach, and L = 29.87 mm and k = 0.48 year1 on Pernambuquinho beach. The growth rate curves for A. brasiliana suggest the presence of three well defined cohorts, thus pointing to a continuous reproduction cycle with peak recruitment between October/2007 and March/2008. A. brasiliana density decreased predominantly during the heavy rains of 2008, probably due to an accumulation of sediments and decreasing salinity. This disturbance probably affected population of this species in the region.


O molusco bivalve Anomalocardia brasiliana há décadas vem sendo coletado em região estuarina do semi-árido do nordeste Brasileiro. No entanto, são escassas as informações sobre a dinâmica e abundância de suas populações na região. Neste contexto, o presente trabalho objetivou estudar aspectos da estrutura populacional de A. brasiliana em praias localizadas próximas à região de estuário do rio Apodi/Mossoró - RN no intuito de verificar a densidade populacional de A. brasiliana durante as diferentes épocas do ano; estimar os parâmetros populacionais e relacionar as características abióticas de região com a distribuição e densidade desta espécie. As coletas foram realizadas no período de março/2007 a maio/2008 nas praias de Barra e Pernambuquinho no município de Grossos - RN. Em cada ponto foram realizados transcectos de 180 metros na região de entremarés e coletadas amostras de sedimento e de moluscos. O maior valor médio de densidade na praia de Pernambuquinho foi de 1148 indivíduos/m2, no mês de abril/07 e o menor foi em maio/08 (100 indivíduos/m2). Na praia de Barra o maior valor foi de 1813 indivíduos/m2, no mês de abril/08 e o menor foi em novembro/07 com 951 indivíduos/m2. Os comprimentos variaram de 1 a 28 mm, sendo mais frequentes indivíduos com comprimentos entre 22 a 24 mm. Foram constatadas diferenças significativas entre as densidades de A. brasiliana em ambas as praias, apenas nos meses de janeiro, abril e maio de 2008. Os parâmetros que descrevem o crescimento em comprimento de A. brasiliana na praia de Barra foram: L = 28,68 mm e k = 0,61 ano1, enquanto que para a praia de Pernambuquinho foram: L = 29,87 mm e k = 0,48 ano1. As curvas de crescimento de A. brasiliana sugerem a presença de três coortes bem definidas, indicando uma reprodução contínua com picos de recrutamento ocorrerendo entre os meses de outubro/07 a março/08. Pode-se verificar uma diminuição da densidade de A. brasiliana, especialmente no período de chuvas intensas do ano de 2008, o que provavelmente esteve associado ao aporte de sedimentos e a diminuição da salinidade. Esta perturbação provavelmente afetou as populações desta espécie na região.

Animal models of gentamicin nephrotoxicity present acute tubular necrosis associated with inflammation, which can contribute to intensify the renal damage. Hydrogen sulfide (H2S) is a signaling molecule involved in inflammation. We evaluated the effect of DL-propargylglycine (PAG), an inhibitor of endogenous H2S formation, on the renal damage induced by gentamicin. Male Wistar rats (N = 8) were injected with 40 mg/kg gentamicin (im) twice a day for 9 days, some of them also received PAG (N = 8, 10 mg·kg-1·day-1, ip). Control rats (N = 6) were treated with saline or PAG only (N = 4). Twenty-four-hour urine samples were collected one day after the end of these treatments, blood samples were collected, the animals were sacrificed, and the kidneys were removed for quantification of H2S formation and histological and immunohistochemical studies. Gentamicin-treated rats presented higher sodium and potassium fractional excretion, increased plasma creatinine [4.06 (3.00; 5.87) mg percent] and urea levels, a greater number of macrophages/monocytes, and a higher score for tubular interstitial lesions [3.50 (3.00; 4.00)] in the renal cortex. These changes were associated with increased H2S formation in the kidneys from gentamicin-treated rats (230.60 ± 38.62 µg·mg protein-1·h-1) compared to control (21.12 ± 1.63) and PAG (11.44 ± 3.08). Treatment with PAG reduced this increase (171.60 ± 18.34), the disturbances in plasma creatinine levels [2.20 (1.92; 4.60) mg percent], macrophage infiltration, and score for tubular interstitial lesions [2.00 (2.00; 3.00)]. However, PAG did not interfere with the increase in fractional sodium excretion provoked by gentamicin. The protective effect of PAG on gentamicin nephrotoxicity was related, at least in part, to decreased H2S formation.

Experimental and clinical evidence suggests that angiotensin II (AII) participates in renal development. Renal AII content is several-fold higher in newborn rats and mice than in adult animals. AII receptors are also expressed in higher amounts in the kidneys of newborn rats. The kidneys of fetuses whose mother received a type 1 AII receptor (AT1) antagonist during gestation present several morphological alterations. Mutations in genes that encode components of the renin-angiotensin system are associated with autosomal recessive renal tubular dysgenesis. Morphological changes were detected in the kidneys of 3-week-old angiotensin-deficient mice. Mitogen-activated protein kinases (MAPKs) are important mediators that transduce extracellular stimuli to intracellular responses. The MAPK family comprises three major subgroups, namely extracellular signal-regulated protein kinase (ERK), c-jun N-terminal kinases (JNK), and p38 MAPK (p38). Important events in renal growth during nephrogenesis such as cellular proliferation and differentiation accompanied by apoptosis on a large scale can be mediated by MAPK pathways. A decrease in glomerulus number was observed in embryos cultured for 48 and 120 h with ERK or p38 inhibitors. Many effects of AII are mediated by MAPK pathways. Treatment with losartan during lactation provoked changes in renal function and structure associated with alterations in AT1 and type 2 AII (AT2) receptors and p-JNK and p-p38 expression in the kidney. Several studies have shown that AII and MAPKs play an important role in renal development. However, the relationship between the effects of AII and MAPK activation on renal development is still unclear.

Mitogen-activated protein kinases (MAPK) may be involved in the pathogenesis of acute renal failure. This study investigated the expression of p-p38 MAPK and nuclear factor kappa B (NF-kappaB) in the renal cortex of rats treated with gentamicin. Twenty rats were injected with gentamicin, 40 mg/kg, im, twice a day for 9 days, 20 with gentamicin + pyrrolidine dithiocarbamate (PDTC, an NF-kappaB inhibitor), 14 with 0.15 M NaCl, im, twice a day for 9 days, and 14 with 0.15 M NaCl , im, twice a day for 9 days and PDTC, 50 mg kg-1 day-1, ip, twice a day for 15 days. The animals were killed 5 and 30 days after the last of the injections and the kidneys were removed for histological, immunohistochemical and Western blot analysis and for nitrate determination. The results of the immunohistochemical study were evaluated by counting the p-p38 MAPK-positive cells per area of renal cortex measuring 0.05 mm². Creatinine was measured by the Jaffé method in blood samples collected 5 and 30 days after the end of the treatments. Gentamicin-treated rats presented a transitory increase in plasma creatinine levels. In addition, animals killed 5 days after the end of gentamicin treatment presented acute tubular necrosis and increased nitrate levels in the renal cortex. Increased expression of p-p38 MAPK and NF-kappaB was also observed in the kidneys from these animals. The animals killed 30 days after gentamicin treatment showed residual areas of interstitial fibrosis in the renal cortex, although the expression of p-p38 MAPK in their kidneys did not differ from control. Treatment with PDTC reduced the functional and structural changes induced by gentamicin as well as the expression of p-p38 MAPK and NF-kappaB. The increased expression of p-p38 MAPK and NF-kappaB observed in these rats suggests that these signaling molecules may be involved in the pathogenesis of tubulointerstitial nephritis induced by gentamicin.

Pemphigus foliaceus (PF) is an autoimmune bullous disease endemic in Brazil. Since serum IL-12 is increased in patients with PF and Langerhans cells (LC) produce IL-12, we titrated serum autoantibodies by indirect immunofluorescence, and quantified epidermal dendritic cells, known as LC, and dermal dendritic cells (DC). Biopsies of blistering lesions were obtained from 22 patients, 13 of whom were submitted to biopsy of both injured and of apparently healthy skin. The control groups consisted of skin from 8 cadavers and from 12 women submitted to breast plastic surgery. LC and DC were identified with anti-CD1a antibody and quantified by morphometric analysis. LC number in the lesion and in apparently healthy skin from PF patients was similar to that of both control groups. DC number in the injured skin (median = 0.94 DC/mm basement membrane) was higher than that of the cadaver group (median = 0.13 DC/mm basement membrane). In the 13 patients with biopsies of both injured and apparently healthy skin, LC and DC were present in larger numbers in the lesion. There was a direct correlation between DC number in the lesion of the PF group and serum autoantibody titers. This correlation was not observed for LC number. The increased number of DC in the lesion, as well as its direct correlation with serum autoantibody titers suggest the participation of DC in the pathogenesis of PF. The relationship between increased DC number and IL-12 in PF needs to be clarified.

The aim of the present study was to investigate the expression of alpha-smooth muscle actin (alpha-SM-actin) and proliferating cell nuclear antigen (PCNA) in renal cortex from patients with focal segmental glomerulosclerosis (FSGS) and their correlations with parameters of renal disease progression. We analyzed renal biopsies from 41 patients with idiopathic FSGS and from 14 control individuals. The alpha-SM-actin immunoreaction was evaluated using a score that reflected the changes in the extent and intensity of staining in the glomerular or cortical area. The PCNA reaction was quantified by counting the labeled cells of the glomeruli or renal cortex. The results, reported as median + or - percentile (25th; 75th), showed that the alpha-SM-actin scores in the glomeruli and tubulointerstitium from the renal cortex were 2.0 (2.0; 4.0) and 3.0 (3.0; 4.0), respectively, in patients with FSGS, and 0.5 (0.0; 1.0) and 0.0 (0.0; 0.5) in the controls. The number of PCNA-positive cells per glomerulus and graded field of tubulointerstitium from the renal cortex was 0.2 (0.0; 0.4) and 1.1 (0.3; 2.2), respectively, for patients with FSGS, and 0.0 (0.0; 0.5) and 0.0 (0.0; 0.0) for controls. The present data showed an increase of alpha-SM-actin and PCNA expression in glomeruli and renal cortex from FSGS patients. The extent of immunoreaction for alpha-SM-actin in the tubulointerstitial area was correlated with the intensity of proteinuria. However, there was no correlation between the kidney expression of these proteins and the reciprocal of plasma creatinine level or renal fibrosis. These findings suggest that the immunohistochemical alterations may be reversible

Antimoniais pentavalentes sao importantes no tratamento da leishmaniose. Seus efeitos mais graves que tem sido relatados sao o aumento do nivel de enzimas hepaticas e anormalidades eletrocardiograficas. Nefrotoxicidade tem sido raramente relatada. Nos relatamos um caso de leishmaniose cutanea generalizada, envolvendo um paciente masculino de 50 anos de idade, que foi submetido ao tratamento com Glucantime. Ele desenvolveu insuficiencia renal devido a necrose tubular aguda e depois veio a obito; apos receber um total de 53 ampolas de Glucantime. O tratamento com o Glucantime foi o responsavel pela necrose tubular aguda diagnosticada em nosso caso

O objetivo do trabalho foi verificar a prevalência de candidíase em transplantados renais. Foram avaliados os prontuários dos pacientes transplantados no Hospital das Clínicas de Ribeirão Preto de fevereiro de 1968 a fevereiro de 1995. Nesse período foram transplantados 620 pacientes. Destes, 87 apresentaram 107 episódios de candidíase. Locais mais acometidos: trato urinário com 55 episódios, trato respiratório com 28, e trato gastrointestinal com 16. O agente etiológico mais freqüente foi C. albicans com 65 casos seguido de C. tropicalis com 12 e C. glabrata com 11 casos. As infecções do trato urinário mostraram incidência maior (61,7%) nos primeiros 6 meses. A maioria se apresentou clínicamente como infecção bacteriana. No trato respiratório, as infecções foram caracterizadas por recuperação do agente no escarro. No trato gastrointestinal, 9/16 episódios foram esofágicos, com epigastralgia, dor retroesternal, às vezes acompanhados de candidíase oral ou odinofagia. Nos outros episódios o agente foi recuperado nas fezes com quadro clínico de gastroenterite. Nas infecções dos tratos urinário e respiratório, houve associação da candidíase com antibioticoterapia prévia (76% e 67% respectivamente), além de infecções bacterianas concomitantes (34% e 64% respectivamente). As infecções por Candida sp tiveram prevalência geral em torno de 14,5%. A localização predominante foi no trato urinário e, em seguida, nos tratos respiratório e gastrointestinal, apresentando alto índice de associação com antibioticoterapia prévia e infecções bacterianas.


The medical records of 620 patients submitted to renal transplant from February 1968 to February 1995 were surveyed for Candida infection. Of these, 87 presented 107 episodes of candidiasis. In 42.9% the infection appeared up to 6 months after the transplant. The most frequent involved sites were: urinary tract, respiratory tract, and gastrointestinal tract. The most frequent etiological agents were: C. albicans, C. tropicalis and C. glabrata. Most urinary tract infections occurred in the first 6 months (61.7%) and manifested clinically as a bacterial infection. In the respiratory tract infections were characterized by isolation of the agent in sputum. In the gastrointestinal infections, 9/16 episodes were esophageal. There were 3 deaths directly related to Candidiasis (one pulmonary and 2 disseminated cases). In the urinary tract, and respiratory tract infections there was association of candidiasis with previous antibiotic treatment (76% and 67%, respectively), and with concomitant bacterial infections (34% and 64%, respectively). The overall prevalence of Candida infections was 14.5%. The predominant location was in the urinary tract (51.0%), followed by the respiratory (26.0%) and gastrointestinal tract (15.0%), with a high rate of association with previous antibiotic treatment and bacterial infections.

Several lines of experimental evidence have shown that transforming growth factor ß (TGF ß) may play a major role in glomerular diseases, mediating the inflammatory response through glomerulosis. In the present study we evaluated TGF ß activity in occasional urine samples from 7 normal individuals and from 15 patients (10 with focal glomerular sclerosis and 5 with membranous glomerulonephritis) using a CCL-64 mink lung cell growth inhibition assay. Urinary TGF ß activity (reported in relation to urine creatinine concentration, Ucr, mean +/- SD) was higher in patients with focal glomerular sclerosis (x/- 17.32 +/- 15.75 / 10 µg Ucr) and patients with membranous glomerulonephritis (x/- = 17.78 +/- 11.53 / 10 µg Ucr) than in normal individuals (x/- = 0.8 +/- 0.44 / 10 µg Ucr). RESUMO

O uso de corticosteróides e/ou outras drogas imunossupressoras no tratamento de nefrite intersticial aguda (NIA) permanece controverso. Os autores relatam um caso de NIA, secundária ao uso de rifampicina, tratado com prednisona, no qual se obteve excelente resposta: paciente feminina com diagnóstico de tuberculose pulmonar, que se apresentava com funçäo renal normal, foi submetida a terapêutica tríplice (rifampicina e pirazinamida). No quadragésimo dia de tratamento, mostrou creatinina de 2,4mg/dl e BUN de 27mg/dl; após três dias, foi submetida a exames que mostraram: creatinina plasmática, 5,5mg/dl; nitrogênio uréico plasmático, 74mg/dl; sódio urinário, 78mEq/1; osmolalidade urinária, 416mOsm/Kg de H2O e fraçäo de excreçäo de sódio de 4,3%; exame do sedimento urinário com leucocitúria, hematúria e eosinofilúria; proteinúria de 24 horas de 900mg; ultra-som renal normal. A biopsia renal revelou tratar-se de NIA, sendo suspensa a rifampicina e, como a funçäo renal continuava a piorar, iniciou-se prednisona. Houve estabilizaçäo dos níveis de escórias nitrogenadas e queda a partir do quarto dia de tratamento, recebendo alta hospitalar com funçäo renal normal

RESUMO

Foram observadas alteraçöes estruturais nos rins de ratos espontaneamente hipertensos (SHR) que antecedem o estado hipertensivo e as alteraçöes hemodinâmicas do nefro. Estes achados podem representar defeito genético estrutural ou estarem associados a hiperatividade autonômica simpática prévia, já observada nos animais jovens pré-hipertensos. Ainda, essas alteraçöes poderiam desempenhar certo papel no desenvolvimento e na manutençäo dos níveis tensionais elevados nesta linhagem de ratos. Os resultados do presente trabalho mostram alteraçöes funcionais peculiares no SHR e confirmam aquelas observadas em animais normotensos adultos após a desnervaçäo renal bilateral
